You can take the AirTrain direct from your terminal atJFK to Jamiaca. There you will get the Long Island Rail Road to Penn Station. At Penn Station you will get an Amtrak train to Wilmington.

You can buy a combined AirTrain/LIRR ticket when you get off the AirTrain in Jamaica, not when you first board at the airport. No reservations are needed.

You will buy a separate Amtrak ticket. You need a reservation for that and you can go ahead and buy your ticket online now. There are Northeast Regional trains and slightly faster Acela trains. The Northeast Regional trains are usually quite a bit cheaper and the time saving with an Acela train to Wilmington will be very small.

If you buy your Amtrak ticket online you will print it out from a kiosk at a QuikTrack machine in the Amtrak portion of Penn Station.

Although I don't think it's worth it in your case, a slightly cheaper alternative would be to take the subway from Jamaica to Penn Station rather than the Long Island Rail Road. You'd take the E train from Jamaica to 34th Street/Penn Station.
